Expansion and Differentiation of H7 Human Embryonic Stem Cells in Chemically Defined Medium Using Corning® Synthemax™ Surface 21 Jun 2010: In this application note Corning Synthemax Surface is used to support long-term self-renewal and differentiation of H7 hESCs. Cells demonstrated a stable doubling time, high level of expression of hESC phenotypic markers (Oct4, SSEA4 and TRA-1-60) and normal karyotype. Corning Synthemax Surface also supported successful directed differentiation of H7 cells into functional cardiomyocytes, proving suitability of Corning Synthemax Surface for both expansion and differentiation of hESC culture. Human Oncostatin M is Functionally Equivalent to Mouse LIF in Supporting Mouse ES Cell Pluripotency & Germline Competency 21 Jun 2010: In this application note R&D Systems demonstrate human OSM is able to maintain ES cell pluripotency in a similar manner to mouse LIF, including long-term maintenance of pluripotency and the ability to differentiate in vitro, the ability to activate STAT3, and germline transmission competency. The study suggests that OSM may be an additional factor that plays a role in early embryonic development and that it may be used as an alternative to LIF in mouse ES cell culture. R&D Systems, Inc.

CO2 Incubator Sterilization method with 90ºC Moist Heat Decontamination 17 Jun 2010: In this application note, Esco validate the Swiftcon 90ºC moist heat decontamination cycle of the Celculture CO2 Incubator by performing decontamination tests on 5 bacterial suspension preparations, including Aspergillus niger and Bacillus subtilis. After decontamination, the microbial suspension discs were placed on tubes containing PBS which were assayed by culturing methods to quantitate Total Viable Count at each location. Esco Micro Pte Ltd

Stem Cell-Derived Human Neurons in CNS Drug Discovery 07 Jun 2010: In this application note from Molecular Devices reproducible batches of human neurons are generated that will significantly improve and impact CNS drug discovery. The stem cell-derived neurons express all known markers typically associated with mature neurons and are functionally active. These neurons have been used in a recent screen to identify antagonists of a novel GPCR target implicated in schizophrenia. Molecular Devices

Optimizing Donor Number for Consistent Pooled Human Liver Microsomes 08 Apr 2010: Human liver microsomes (HLMs) are commonly used as an in vitro reagent for the study of human CYP in the metabolism of a drug candidate. This application note presents an analysis of the CYP activity distributions for over 300 characterized (HLM) samples and predicts the variability in pooled CYP activity as a function of donor number. An optimal donor number based on observed variability can be selected. BD Biosciences

HTRF® Solutions for Oncology 04 Feb 2010: This application note presents a comprehensive line of HTRF (homogeneous time-resolved fluorescence) reagents for assessing oncology-related targets such kinases, ATPases, ubiquitin and heparanase assays. In addition, HTRF reagents are available for studying cell signaling, biomarker discovery, ligand binding, receptor and second messenger. Cisbio Bioassays

A Flow Cytometer with Fixed Voltage PMTs and Expanded Dynamic Range Simplifies Collection and Analysis of Complex Cell Populations 13 Mar 2009: Due to the wide variations in fluorescence and light scatter signals produced by different types of cells and particles, flow cytometrists often spend significant time, and waste significant amounts of sample, optimizing amplifier gains and/or PMT voltage settings. We demonstrate here that a flow cytometer equipped with fixed voltage PMTs calibrated to operate in linear range, can be used to analyze a wide variety of sample sizes. Accuri Cytometers, Inc.

AFP Assay on Olympus AU3000i Immunoassay Analyser
08 Jul 2008: The AU3000i is a new immunoassay analyser from Olympus Diagnostics. It is a high throughput analyser, using the same sample racks as the Olympus family of general chemistry analysers (AU640, 2700, etc). This poster presents an assessment of the AFP assay as conducted in a District General Hospital in the North East of England Olympus Europa Holding GmbH
